Product Specific Information

The monoclonal antibody Moushi101 recognizes mouse CD101 (IGSF2, V7), a transmembrane glycoprotein with 7 IgV domains and phosphorylation sites. CD101 is expressed on monocytes, granulocytes, dendritic cells, Langerhans cells, and activated T cells. It is also found on CD25+Foxp3+ T regulatory cells with high suppressive activity.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes, ≤0.25 µg/test in 100 µL final volume. Optimal titration recommended.

Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Strain Note:
NovaFluor Yellow 730 shows non-specific staining of B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD101 (V7 antigen) is a disulfide-linked homodimer of the Ig superfamily expressed on monocytes, granulocytes, dendritic cells, and T lymphocytes. It plays a role in T-cell activation and proliferation. In mice, CD101 is found on regulatory T cells (CD4+, CD25+, Foxp3+) with high suppressor activity. In human PBMCs, CD101 is expressed on ~30% of Foxp3+ cells, though enhanced suppressor activity has not been confirmed.
