Product Specific Information

The LN3 monoclonal antibody reacts with the human major histocompatibility complex (MHC) class II molecule, HLA-DR.
HLA-DR is expressed on the surface of antigen-presenting cells (APCs) such as B cells, monocytes, macrophages, dendritic cells, and activated T cells.
It is a heterodimeric transmembrane protein composed of alpha and beta subunits, playing a key role in peptide presentation to CD4⁺ T lymphocytes.

  • Applications Reported: Flow cytometric analysis
  • Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells
  • Recommended Use: 5 µL (0.125 µg) per test in a final 100 µL staining volume
  • Typical Cell Range: 10⁵–10⁸ cells/test

Brilliant Violet™ 421 (BV421) emits at 423 nm and is optimized for cytometers with a 405 nm violet laser.
For panels using multiple polymer dyes (Super Bright, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

HLA-DR is a transmembrane glycoprotein composed of a 36 kDa alpha chain (DRA) and a 27 kDa beta chain (DRB).

  • The DRA gene contains 5 exons encoding the leader peptide, extracellular domains, transmembrane region, and cytoplasmic tail.
  • DRA acts as the sole alpha chain for DRB1, DRB3, DRB4, and DRB5, with DRB1 containing the polymorphisms that determine peptide-binding specificity.
  • HLA-DR is expressed mainly on B lymphocytes, monocytes, macrophages, thymic epithelial cells, and activated T lymphocytes.
  • The DR, DQ, and DP loci encode the major expressed products of the human MHC class II region.

These molecules bind processed peptides and present them to T-helper cells, initiating immune responses.
HLA and MHC antibodies are essential tools in Immunopeptidomics, aiding in neoantigen identification and characterization via LC-MS/MS.
