Product Specific Information

The monoclonal antibody TWAJ recognizes mouse and human Gata-3, a member of the Gata family of transcription factors.
Gata-3 is a T cell-specific transcription factor critical for thymic development and Th2 differentiation.
During embryonic development, Gata-3 expression is found in the central nervous system, skin, mammary glands, and kidney, and is essential for survival—homozygous knockout is embryonic lethal.

In the thymus, Gata-3 is mainly expressed on CD4 single-positive cells.
During Th2 differentiation, Gata-3 binds to the IL-4 promoter and represses T-bet expression, inhibiting Th1 differentiation.

Alternative splice variants (50 and 45 kDa) have been reported, particularly in the MCF7 cell line.
The TWAJ antibody recognizes both splice forms.

Staining Requirement: Use the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00).
Crossreactivity: Published for rhesus monkey.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-diluted and validated for intracellular staining of mouse thymocytes using the Foxp3/Transcription Factor Staining Buffer Set.

A test is defined as the amount (µg) of antibody that stains a cell sample in a final volume of 100 µL (typically 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 421 (BV421) emits at 423 nm and is optimized for cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Target Information

GATA3 regulates transcription of all four T-cell receptor subunit genes (α, β, γ, δ) through enhancer binding.
It contains zinc finger domains and is expressed specifically in T cells.
GATA3 mRNA is abundant in T-lymphocytes but absent in B cells and macrophages.
The human gene spans 17 kb with six exons; its zinc fingers are conserved with GATA1.
GATA3 plays a key role in T-cell receptor gene activation and enhancer binding.
