Product Specific Information

The B3B4 monoclonal antibody reacts with mouse CD23, a 45 kDa type II transmembrane glycoprotein expressed on resting conventional B cells. Its expression is modulated upon B-cell activation and is absent in B-1 cell lineage (CD5+ B cells). Soluble CD23 forms are biologically active and function as low-affinity receptors for IgE, playing roles in IgE response regulation and B-cell activation. CD21 is thought to bind to CD23.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes; recommended ≤0.4 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL of CellBlox Plus per sample (regardless of the number of NovaFluors).
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, employing novel nucleic acid dye structures engineered for optimal fluorescence with minimal spillover and spread.

Target Information

CD23 is a 45 kDa glycoprotein expressed on a subset of peripheral blood and tonsil B cells, and strongly on EBV-transformed B lymphoblasts. It functions as a low-affinity IgE receptor and is involved in B-cell growth, differentiation, and IgE production. Soluble CD23 acts as a potent mitogenic factor. Abnormal CD23 expression is associated with conditions such as chronic conjunctivitis and chronic lymphocytic leukemia.
