Product Specific Information

The B3B4 monoclonal antibody reacts with mouse CD23, a 45 kDa type II transmembrane glycoprotein expressed on resting conventional B cells and modulated upon activation. B-1 lineage cells (CD5+ B cells) do not express CD23. Soluble CD23 is biologically active and serves as a low-affinity receptor for IgE, involved in IgE response regulation and B-cell activation. CD21 is known to bind CD23.

• Applications Tested: Tested on mouse splenocytes, ≤0.4 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titer for best results.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains with NovaFluor dyes.
• Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
  • Add 5 µL per sample before combining with cells.
  • For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescence signatures with minimal spillover and spread.

Note: Internal testing shows NovaFluor Yellow 610 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 strains.

Target Information

CD23 is a 45 kDa glycoprotein expressed on a subset of peripheral blood and tonsil B cells, and strongly on EBV-transformed B lymphoblasts. It functions as the low-affinity IgE receptor (FcεRII) and plays roles in B-cell growth, differentiation, and IgE regulation. Soluble CD23 acts as a potent mitogenic factor. Abnormal CD23 expression is associated with chronic lymphocytic leukemia and certain lymphomas.
