Product Specific Information
The B3B4 monoclonal antibody reacts with mouse CD23, a 45 kDa type II transmembrane glycoprotein.
CD23 is expressed on resting conventional B cells and modulated upon B-cell activation. B-1 lineage (CD5+ B cells) does not express CD23. Soluble CD23 forms are biologically active and function as a low-affinity receptor for IgE, regulating IgE response and B-cell activation. CD21 is known to bind CD23.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes using ≤0.4 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.
Each product includes:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Important Usage Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
- Add 5 µL of CellBlox Plus Blocking Buffer per sample (regardless of number of NovaFluors) before combining with cells.
- For single-color controls: use 5 µL buffer per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures, enabling optimized fluorescent signatures with reduced spillover and spread.

Target Information
CD23 is a 45 kDa glycoprotein expressed on a subset of peripheral blood and tonsil B cells, and strongly expressed on EBV-transformed B lymphoblasts. It serves as the low-affinity IgE receptor and is involved in B-cell growth, differentiation, and IgE production. Soluble CD23 acts as a potent mitogenic factor.
CD23 dysfunction is associated with diseases such as chronic conjunctivitis and chronic lymphocytic leukemia.
