Product Specific Information
Description:
The B3B4 monoclonal antibody reacts with mouse CD23, a 45 kDa type II transmembrane glycoprotein expressed on resting conventional B cells. Its expression changes upon B-cell activation, while B-1 lineage (CD5+ B cells) does not express CD23. CD23 acts as a low-affinity receptor for IgE, involved in IgE regulation and B-cell activation. CD21 is known to bind CD23.
Applications Reported:
Suitable for flow cytometric analysis.
Applications Tested:
Tested by flow cytometry using mouse splenocytes. Recommended ≤0.4 µg per test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
- Add 5 µL per sample (regardless of number of NovaFluors).
- For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information
CD23 is a 45 kDa glycoprotein present on a subset of peripheral blood and tonsil B cells, strongly expressed on EBV-transformed B lymphoblasts. It functions as the low-affinity IgE receptor on B cells and is involved in B-cell growth, differentiation, and IgE regulation. Soluble CD23 acts as a mitogenic factor. Dysregulation is associated with conditions such as chronic conjunctivitis and chronic lymphocytic leukemia.
