Description:
The TRA-1-81 antibody recognizes a protein expressed on undifferentiated human embryonic stem cells (ES), embryonic carcinoma cells (EC), and embryonic germ cells (EG). The epitope is lost upon differentiation and is resistant to neuraminidase digestion, unlike TRA-1-60. It specifically binds a carbohydrate epitope on podocalyxin, a CD34-related sialomucin found on podocytes and associated with cancer aggressiveness (e.g., breast, prostate cancer).

Flow cytometry, immunoprecipitation, immunoblotting, immunofluorescence.
Applications Tested:
- Flow cytometry: ≤0.5 µg/test (2102Ep cell line)
- Immunoblotting: Band at 200–250 kDa (reducing/non-reducing)
- Immunofluorescence: ≤5 µg/mL (formaldehyde-fixed, permeabilized cells)

Target Information
TRA-1-81 is a cell surface antigen expressed with SSEA-3, SSEA-4, and TRA-1-60 in human embryonic stem cells, embryonal carcinoma cells, and induced pluripotent stem cells (iPS). These markers are down-regulated during differentiation, whereas SSEA-1 appears after retinoic acid-induced differentiation.
