Product Specific Information
The eBioABL-93 monoclonal antibody reacts with mouse CD107b, also known as lysosomal-associated membrane protein-2 (LAMP-2). CD107b is a highly glycosylated protein (~120 kDa) expressed intracellularly in lysosomal membranes. It is transiently expressed on the surface of cytolytic T cells during degranulation, though less than CD107a, and may be identical to the mouse Mac-3 antigen.

- Intracellular staining followed by flow cytometric analysis
- Surface staining in flow cytometric degranulation assays
Applications Tested:
This antibody has been tested by intracellular staining and flow cytometric analysis of thioglycolate-elicited peritoneal exudate cells. It can be used at ≤0.5 µg per test (defined as the amount that stains a cell sample in 100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Antibody titration is recommended for optimal performance.

Target Information
LAMP2 (Lysosome-associated membrane glycoprotein 2, CD107b) is a member of a family of membrane glycoproteins. It provides selectins with carbohydrate ligands and may play roles in tumor metastasis, lysosomal protection, and membrane adhesion. The unmodified LAMP2 polypeptide is approximately 46 kDa, while mature glycosylated forms range from 100–130 kDa. Structurally, LAMP2 consists of:
- A large amino-terminal intra-lysosomal domain
- A hydrophobic transmembrane domain
- A short cytoplasmic tail
LAMP2 participates in chaperone-mediated autophagy and lysosome-phagosome fusion. It exists in three variants (LAMP-2A, -2B, -2C). LAMP2 deficiency leads to autophagosome accumulation and is associated with Danon’s disease.
