Product Specific Information

The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated protein (~110 kDa) predominantly expressed in lysosomal/endosomal membranes. CD107a transiently appears on the surface of degranulating cytolytic T cells and is upregulated on activated platelets and certain cancer cells.

Applications Reported:

  • Intracellular staining followed by flow cytometric analysis
  • Surface staining in flow cytometric degranulation assays

Applications Tested:
Pre-titrated and validated for intracellular staining and flow cytometric analysis of Jurkat cells.
Use at 5 µL (0.5 µg) per test (final volume 100 µL). Cell number per test: 10⁵–10⁸ cells.

Target Information

LAMP1 (CD107a) is a major lysosomal membrane component alongside LAMP2. About 1–2% of CD107a is found on the plasma membrane. It is a type I integral membrane protein with a signal peptide, 18 N-linked glycosylation sites, a single transmembrane segment, and a short cytosolic tail.

Biological Role:

  • Involved in lysosome biogenesis and fusion with phagosomes
  • Transports from trans-Golgi network to endosomes and lysosomes
  • Surface expression upon cell activation depends on a carboxyl-terminal tyrosine motif (YXXI)
  • Mutations in this motif affect lysosomal localization and membrane cycling
  • Promotes PBMC adhesion to vascular endothelium, suggesting a role in inflammation
  • Increased expression in neurons and glial cells near senile plaques in Alzheimer’s disease
  • Marker of mast cell activation
