Product Specific Information

The CBRM1/5 monoclonal antibody reacts with an activation-specific epitope of human Mac-1. It binds a subset of Mac-1 molecules on neutrophils and monocytes after stimulation with chemoattractants or phorbol esters but does not recognize Mac-1 on resting myeloid cells.
Mac-1 participates in adhesive cell interactions via its ligands. The epitope recognized by this antibody localizes to the I domain on the alpha chain of Mac-1 near the ligand binding site. CBRM1/5 blocks Mac-1–dependent adhesion to fibrinogen and ICAM-1 and inhibits chemoattractant-stimulated adhesion of eosinophils to ICAM-1.

Note: Low-level activation may occur during processing of freshly drawn blood, possibly leading to some binding to Mac-1 even in unstimulated samples. Higher levels are observed in activated samples.

Applications Tested: Pre-titrated and tested on resting and activated human peripheral blood leukocytes at 5 µL (0.5 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.

Fluorochrome Information:

  • Alexa Fluor® 700 emits at 723 nm and is excited with the red laser (633 nm).
  • Recommended filter: 685 LP mirror and 710/20 filter.
  • Excitation: 633–647 nm; Emission: 723 nm; Laser: Red Laser.

Target Information

CD11b (integrin alpha-M, ITGAM, integrin alpha-X, ITGAX) is a 165 kDa adhesion molecule that associates non-covalently with integrin beta-2 (CD18).
The CD11b/CD18 complex (Mac-1, CR3) is expressed on monocytes/macrophages, granulocytes, activated lymphocytes, subsets of NK and dendritic cells, and microglia.
It serves as a receptor for ICAM-1, ICAM-2, ICAM-4, CD14, CD50, CD23, heparin, iC3b, fibrinogen, and Factor X, mediating critical cell-cell and cell-matrix interactions.

Expression levels:

  • 8% of spleen cells
  • 44% of bone marrow cells
  • <1% of thymocytes

CD11b is commonly used as a microglial marker. Expression increases during monocyte maturation and varies among tissue macrophages, with peritoneal macrophages showing higher expression than splenic macrophages.

Diseases associated with CD11b dysfunction include systemic lupus erythematosus and ITGAM-related susceptibility to autoimmune disorders.
