Amino Acid Sequence of Recombinant Protein:

Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human MIP-1β (CCL4). The antibody was purified by affinity chromatography and subsequently biotinylated.

Sandwich ELISA:
To detect Human MIP-1β by sandwich ELISA (100 µL/well antibody solution), a concentration of 0.25–1.0 µg/mL is required. This biotinylated polyclonal antibody, together with PeproTech Polyclonal Anti-Human MIP-1β (500-P38B) as a capture antibody, allows detection of at least 0.2–0.4 ng/well of Recombinant Human MIP-1β.

Western Blot:
For detection of Human MIP-1β by Western Blot, use at 0.1–0.2 µg/mL. When used with compatible secondary reagents, the detection limit for Recombinant Human MIP-1β is 1.5–3.0 ng/lane under reducing or non-reducing conditions.

Target Information

CCL4 (Macrophage Inflammatory Protein 1-beta, MIP-1β) belongs to the intercrine beta (chemokine CC) family. It is involved in chemotactic and proinflammatory responses and immune homeostasis. CCL4 is produced by macrophages upon stimulation by bacterial endotoxins and recruits various inflammatory cells to sites of inflammation.

CCL4 and the related protein CCL3 regulate the trafficking and activation of inflammatory cells during immune responses to pathogens. While both affect monocytes similarly, CCL4 selectively attracts CD4+ lymphocytes and CCL3 attracts CD8+ lymphocytes. Both are potent chemoattractants for B cells, eosinophils, and dendritic cells. The processed form of CCL4 can down-modulate CCR5 receptor expression, inhibiting CCR5-mediated HIV-1 entry into T cells. CCL4 binds with high affinity to CCR5 receptors.
