Preparation:
Produced from sera of rabbits immunized with highly pure Recombinant Human MIP-1beta (CCL4). Anti-Human MIP-1beta (CCL4)-specific antibody was purified by affinity chromatography employing an immobilized Human MIP-1beta (CCL4) matrix.
Sandwich ELISA:
For detection of Human MIP-1beta (CCL4) by sandwich ELISA, use 0.5–2.0 µg/mL antibody concentration (100 µL/well). In conjunction with PeproTech Biotinylated Anti-Human MIP-1beta (CCL4) (500-P38BBt) as detection antibody, allows detection of 0.2–0.4 ng/well recombinant protein.
Western Blot:
To detect Human MIP-1beta by Western Blot, use 0.1–0.2 µg/mL antibody. Detection limit for recombinant protein is 1.5–3.0 ng/lane under reducing or non-reducing conditions.
Target Information
CCL4 (macrophage inflammatory protein 1-beta, MIP1B) belongs to the chemokine CC family. It has chemotactic and proinflammatory roles and is produced by macrophages upon stimulation by bacterial endotoxins.
CCL4 recruits inflammatory cells to sites of inflammation and is produced by lymphocytes, macrophages, and dendritic cells.
CCL4 and CCL3 regulate immune responses to pathogens by controlling trafficking and activation of inflammatory cells.
CCL4 selectively attracts CD4+ lymphocytes, while CCL3 attracts CD8+ lymphocytes.
Both act as chemoattractants for B cells, eosinophils, and dendritic cells.
Processed CCL4 can down-modulate CCR5 receptor expression, inhibiting CCR5-mediated HIV-1 entry into T cells.
CCL4 binds with high affinity to CCR5 receptors.
