Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid cycles of freezing and thawing. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

Anti-EBI-3 / IL27 beta Antibody Peroxidase Conjugate is an IgG fraction antibody purified from monospecific antiserum by a multi-step process including delipidation, salt fractionation, and ion-exchange chromatography followed by extensive dialysis against the buffer stated above. This antibody is specific for mouse and rat EBI-3 protein. Cross-reactivity does not occur with human EBI-3. Cross-reactivity with IL-27 from other sources has not been determined.

Target Information

EBI3 (Epstein-Barr virus Induced-3) is a secreted glycoprotein of the hematopoietin receptor family. It plays a critical regulatory role in the induction of Th2-type immune responses and the development of Th2-mediated tissue inflammation in vivo, possibly through control of iNKT cell function.
EBI3 dimerizes with p28 and p35 subunits of IL-12 to form IL-27 and IL-35, respectively. IL-27 is produced early by activated antigen-presenting cells upon TLR ligation and negatively regulates Th17 cell differentiation.
EBI3 is widely expressed and transcriptionally regulated by TLR signaling via MyD88 and NF-kappaB in dendritic cells. It inhibits delayed-type hypersensitivity responses by suppressing IL-17 and inducing IL-10 hyperproduction.
EBI3 may play a role in controlling tumor metastasis via lung CD8+ T cells, and its deficiency is associated with reduced Th2 cytokine production involved in allergic airway inflammation. Targeted deletion of EBI3 protects mice from lung metastasis.
