Product Specific Information

This NGZB monoclonal antibody reacts with mouse Granzyme B, a member of the granzyme serine protease family found in cytotoxic T cells and NK cells. Granzyme B (also known as CGL1 or CTLA-1) is crucial for inducing apoptosis in target cells via the mannose 6-phosphate receptor.
This antibody does not cross-react with human Granzyme B and recognizes a distinct epitope compared to GB11.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Verified using mouse splenocytes with Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00). Recommended ≤1.0 µg/test (100 µL volume, 10⁵–10⁸ cells/test).
Careful titration is advised for optimal performance.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) for up to 3 days at 4°C in the dark.

Excitation/Emission: 488–561 nm / 695 nm
Laser Compatibility: Blue, Green, Yellow-Green lasers.

Target Information

Granzyme B is a serine protease found in cytotoxic T lymphocytes (CTLs) and natural killer (NK) cells. It induces apoptosis through caspase-3 activation and mitochondrial pathways (Bid protein cleavage).
Granzyme B functions via mannose 6-phosphate receptor-mediated uptake and is essential for immune-mediated cytotoxicity. Multiple granzymes (A–G) are cloned from mouse CTLs and NK cells, while fewer are identified in humans.
