• For IHC (PFA-fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Interleukin 10 (IL-10) encodes a cytokine primarily produced by monocytes and some lymphocytes. It plays a crucial role in immunoregulation and inflammation by down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. IL-10 enhances B cell survival, proliferation, and antibody production, and can inhibit NF-kappa B activity while modulating the JAK-STAT signaling pathway.
Knockout studies indicate its essential role in intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis. Associated diseases include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1 infection.
