• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

IL-10 encodes a cytokine primarily produced by monocytes and some lymphocytes. It plays a key role in immunoregulation and inflammation by:

• Down-regulating Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages
• Enhancing B cell survival, proliferation, and antibody production
• Blocking NF-kappa B activity and regulating the JAK-STAT signaling pathway

Knockout studies in mice indicate IL-10 is essential for intestinal immune regulation. Mutations in IL10 are linked to increased susceptibility to HIV-1 infection and rheumatoid arthritis.
Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
