Target Information
KLRB1 (Killer cell lectin like receptor subfamily B, member 1, CD161) protein is a type II membrane protein with an external C terminus. It is expressed by almost all NK cells and a subset of CD3+ T cells. KLRB1 is a homodimeric surface protein (40–44 kDa) that plays an inhibitory role on NK cells. Activation leads to stimulation of acid sphingomyelnase/SMPD1 and increased intracellular ceramide, as well as activation of AKT1/PKB and RPS6KA1/RSK1 kinases, and T cell proliferation by anti-CD3.
KLRB1 acts as a lectin binding to Gal-α(1,3)Gal and N-acetyllactosamine epitopes, and also interacts with CLEC2D/LLT1 as a ligand to inhibit NK cell-mediated cytotoxicity and interferon-γ secretion. The protein contains an extracellular domain with C-type lectin motifs, a transmembrane domain, and a cytoplasmic domain.
