Product Specific Information

Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes followed by cooling at room temperature for 20 minutes.
Following antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is lung carcinoma or placenta tissue.

Target Information

EGFR (epidermal growth factor receptor) is a receptor tyrosine kinase that signals in response to various growth factors. Overexpression has been linked to numerous types of cancer, and EGFR is the target of both biological and small molecule therapeutics.
EGFR is encoded by the EGFR gene located on chromosome 7 in humans and belongs to the HER/ERbB family of proteins, which includes ERbB2, ERbB3, and ERbB4.
Binding of ligands such as EGF and TGF-α to the extracellular domain leads to EGFR dimerization and autophosphorylation of tyrosine residues in the cytoplasmic domain.
Overexpression is observed in tumors of the head and neck, brain, bladder, stomach, breast, lung, endometrium, cervix, vulva, ovary, esophagus, and in squamous cell carcinoma.
