Product Specific Information

The IP26 monoclonal antibody reacts with the alpha-beta chain of human TCR, expressed by the majority of peripheral T cells.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of human peripheral blood cells.
Recommended usage: 5 µL (1 µg) per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test.
Note: Staining may be obstructed when OKT3 (anti-human CD3) antibody is used as a co-stain.
Excitation: 488 nm; Emission: 520 nm; Laser: Blue Laser.
Filtration: 0.2 µm post-manufacturing filtered.

The T cell antigen receptor (TCR) is a ligand-specific α/β heterodimer associated with invariant CD3 γ/δ/η and ζ subunits, essential for surface expression and signal transduction.
TCR activation induces differentiation, proliferation, and cytokine production.
Most peripheral T cells express α/β TCRs, while γ/δ TCRs are mainly found in epithelial tissues.
Proper assembly of TCR-CD3 complexes is required for surface expression; absence of any subunit halts expression.
TCR recognition of self-peptides is linked to autoimmune diseases and tumor-associated mutations.
Superantigen engagement of TCRs is implicated in toxic shock syndrome.
Autoantibodies to Vβ segments are found in RA and SLE patients, potentially modulating inflammatory responses.
Human T cell subsets include CD4⁺, CD8⁺ (single positive), and CD4⁻CD8⁻ (double negative) populations, with altered distributions in autoimmune and immunodeficiency conditions.
γ/δ T cells exhibit limited diversity and distinct antigen recognition, contributing to antitumor and immunoregulatory activity.
