Product Specific Information
The J43 monoclonal antibody reacts with mouse PD-1 (programmed death-1), a 55 kDa member of the Ig superfamily. PD-1 contains the immunoreceptor tyrosine-based inhibitory motif (ITIM) and plays a key role in peripheral tolerance and autoimmune disease in mice. PD-1 is expressed mainly on activated T and B lymphocytes.
Two novel B7 Family members, PD-L1 (B7-H1) and PD-L2 (B7-DC), are identified as PD-1 ligands. J43 inhibits the binding of mouse PD-L1-Ig and PD-L2-Ig to PD-1/BHK transfected cells. In vivo administration of J43 enhances contact hypersensitivity and exacerbates acute GVHD, EAE, and NOD diabetes similar to PD-1-deficient cells.

Target Information
PD-1 (PDCD-1) mediates inhibitory signals in activated T cells when bound to ligands PD-L1 or PD-L2 on antigen-presenting cells. These interactions suppress immune activation in the absence of danger signals such as LPS. PD-1-deficient mice exhibit hyperactivated immune responses and autoimmune disease. Despite its predicted molecular weight, PD-1 often migrates higher in SDS-PAGE.
