Product Specific Information

The dG9 antibody clone reacts with human perforin (pore-forming protein, pfp). Perforin is a cytolytic mediator in cytotoxic T lymphocytes (CTL) and natural killer (NK) cells, essential for immune defense against tumors and virus infections.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis
• Immunohistochemical staining

Applications Tested:

• Intracellular flow cytometric analysis of normal human peripheral blood cells
• Recommended use: 5 µL (0.125 µg) per test (100 µL final volume)
• Cell number range: 10⁵–10⁸ cells/test

Target Information

Perforin is a major cytolytic protein stored in cytoplasmic granules and released by cytotoxic lymphocytes. It plays a key role in immune defense against tumors and viral infections.

• Length: 555 amino acids (including 21 aa signal peptide)
• Molecular Weight: 70–75 kDa
• Mechanism: Pore-forming, similar to complement component C9
• Expression: Found in killer cell lines, not in helper T cells or other tumor cells
• Clinical relevance: Mutations cause familial hemophagocytic lymphohistiocytosis type 2 (HPLH2)
• Alternative splicing results in multiple transcript variants
