Description:
The JS-83 monoclonal antibody reacts with human CD45RA, a 220 kDa molecule expressed by subpopulations of CD4⁺ and CD8⁺ peripheral T lymphocytes, as well as B cells. CD45RA⁺ T cell populations are mainly naïve/virgin, allowing JS-83 to serve as a phenotypic marker to discriminate T cell subsets.

Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.125 µg) per test. A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell numbers may vary from 10⁵ to 10⁸ cells/test.

Target Information

CD45RA is an isoform of the CD45 complex with restricted expression among lymphoid subtypes. CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. It plays a critical role in lymphocyte development and antigen signaling by regulating Src-family kinases.

CD45 isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions. Expression varies by cell type and physiological state. CD45 also modulates integrin-mediated signaling and participates in apoptosis-related DNA fragmentation. It is expressed on hematopoietic cells except mature red cells and absent on non-hematopoietic tissues.

Diseases associated with CD45 dysfunction include severe combined immunodeficiency and Hepatitis C.
