Target Information

Synaptotagmin 1 is a synaptic vesicle membrane glycoprotein widely expressed throughout the central nervous system (CNS). It functions as a Ca²⁺ sensor in the regulation of exocytosis and neurotransmitter release.
Studies indicate that Ca²⁺-mediated binding of Synaptotagmin 1 to SNAP25 is essential for Ca²⁺-dependent membrane fusion. Specific residues within the C2B binding domain independently influence endocytic rate and synaptic vesicle size.
Synaptotagmin 1 may regulate membrane interactions during synaptic vesicle trafficking at the active zone. It binds acidic phospholipids requiring both an acidic head group and a diacyl backbone.
Ca²⁺-dependent interactions occur with receptors for activated protein kinase C, while Ca²⁺-independent interactions occur with neurexins, syntaxin, and AP2.
Dysfunction of Synaptotagmin 1 is associated with food-borne and infant botulism.
