The EM92 monoclonal antibody reacts with mouse and human Oct3/4, encoded by the Pou5F1 gene. Oct3/4 is a POU domain-containing transcription factor critical for maintaining embryonic stem (ES) and induced pluripotent stem (iPS) cells in a pluripotent state. It is expressed by ES cells, embryonic germ cells, and embryonic carcinoma cell lines.
Reduction of Oct3/4 expression in inner cell mass leads to dedifferentiation to trophoectoderm, while increased expression induces differentiation to mesoderm and primitive endoderm. Oct3/4 regulates genes such as FGF-4, UTF1, Sox2, Fbx15, Rex1, and osteopontin, and frequently acts synergistically with Sox2.
Oct3/4 expression can be negatively regulated by retinoic acid treatment or removal of leukemia-inhibitory factor (LIF).

Recommended usage: ≤0.5 µg per test (100 µL final volume). Cell number may vary from 10⁵–10⁸ cells/test. Antibody titration is advised for optimal performance.

POU5F1 (Oct-4) is a maternally expressed octamer-binding transcription factor crucial in early developmental stages. It plays a key role in maintaining pluripotency and has applications in stem cell generation and regenerative medicine.
Artificial stem cells (iPS cells) can be created by expressing POU5F1 along with Sox2, Klf4, and c-Myc in fibroblasts. Recent studies show iPS cells can be generated via plasmid expression without viral introduction, improving safety for therapeutic use.
