Product Specific Information

The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein of the Siglec family. It is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

• Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.25 µg) per test (100 µL final volume).
  Cell number range: 10⁵–10⁸ cells/test.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Target Information

CD33 is a transmembrane protein of the Siglec family containing ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It binds sialic acid residues in N- and O-glycans, and is a therapeutic target for acute myeloid leukemia (AML). CD33 is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors, and peripheral monocytes, but not on pluripotent stem cells. It is useful for distinguishing myelogenous leukemia from lymphoid or erythroid leukemias. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
