Product Specific Information

The WM59 monoclonal antibody reacts with human CD31 (platelet-endothelial cell adhesion molecule-1, PECAM-1, gpIIa), a 130–140 kDa surface protein expressed by endothelial cells and at low levels on leukocytes and platelets. CD31 plays a role in adhesion, cell-cell and cell-matrix interaction, and signal transduction.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on normal human peripheral blood cells at ≤5 µL (0.06 µg)/test. A test is defined as staining a cell sample in 100 µL final volume. Cell number may range from 10⁵–10⁸ cells/test. Optimize titration for best results.

Fluorochrome Information:
Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended detection filter: 660/20 bandpass.

Usage Recommendations:

• Use Super Bright Complete Staining Buffer (Product # SB-4401) when combining multiple Super Bright dyes to reduce non-specific interactions.
• Protect vial and stained samples from light (light-sensitive dye).
• Samples can be fixed with IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C with minimal brightness loss.

Target Information

CD31 (PECAM-1) is an inhibitory coreceptor regulating T and B cell signaling via ITIM motifs. It is expressed throughout the vascular compartment, mainly at intercellular junctions. Its N-terminal Ig-like domain mediates homophilic binding, crucial for cell-cell interactions. CD31 functions in integrin-mediated adhesion, transendothelial migration, angiogenesis, apoptosis, immune regulation, macrophage phagocytosis, IgE-mediated anaphylaxis, and thrombosis.
