Product Specific Information

The 5C3 monoclonal antibody reacts with human CD40, a 45–50 kDa type I transmembrane glycoprotein and member of the TNFR family. CD40 is expressed by B lymphocytes, follicular dendritic cells, thymic epithelium, and a subset of peripheral T cells. It regulates B cell development and maturation by inducing Ig isotype-switching and, together with IL-4, protects B cells from apoptosis and promotes proliferation. Interaction with CD154 (gp39) on T cells is crucial for T-B cell communication and immune regulation.

Fluorochrome Information:
Super Bright 645 is excited with a 405 nm violet laser and emits at 645 nm. Recommended bandpass filter: 660/20. Ensure instrument compatibility.

Staining Buffer Recommendation:
Use Super Bright Complete Staining Buffer (Product # SB-4401) when using multiple Super Bright-conjugated antibodies to minimize non-specific interactions.

Target Information

CD40 is a single-chain glycoprotein belonging to the TNF/NGF receptor superfamily, homologous to CD30. It is present on all B cells except plasma cells, and on some epithelial cells, carcinomas, and lymphoid dendritic cells. CD40 is essential for immune and inflammatory responses, including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation. It is regulated by the AT-hook transcription factor AKNA and interacts with adaptor protein TNFR2. CD40-CD40L interaction is implicated in amyloid-beta-induced microglial activation, relevant to Alzheimer’s disease. Two alternatively spliced transcript variants encoding distinct isoforms have been reported.
