Target Information

The androgen receptor (AR) is a member of the steroid-hormone receptor superfamily of nuclear receptors. It has three major functional domains:

  • N-terminal domain
  • DNA-binding domain
  • Androgen-binding domain

AR acts as a ligand-activated transcription factor binding testosterone (T) and dihydrotestosterone (DHT). Upon ligand binding, AR dissociates from accessory proteins, translocates into the nucleus, dimerizes, and activates transcription of androgen-responsive genes.

The AR signaling pathway is critical for the development and function of male reproductive organs (prostate, epididymis) and also influences nonreproductive tissues such as muscle, hair follicles, and brain. AR is a phosphoprotein that regulates MAP kinase signaling; inhibition of the MEK1/2 pathway affects AR phosphorylation.

Abnormalities in AR signaling are linked to diseases including prostate cancer, Kennedy’s disease, and male infertility. Mutations in the AR gene are associated with complete androgen insensitivity syndrome (CAIS).
