Product Specific Information

eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell surface protein with ecto-5'-nucleotidase activity. Expression on myeloid cells (CD11b) is restricted to the bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling. Differences between human and mouse CD73 expression patterns are noted.

CD73 is expressed on subsets of lymphocytes and increases during maturation. It is found on memory CD4 T cells and regulatory T cells.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis.
Applications Tested: Flow cytometric analysis of mouse splenocytes, ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize titration for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
  • Add 5 µL per sample to antibody cocktails before combining with cells.
  • For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD73 (Ecto-5'-nucleotidase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, preferring AMP as substrate. It forms a dimer of two identical 70 kDa subunits bound to the plasma membrane via GPI linkage. CD73 serves as a marker of lymphocyte differentiation, and its deficiency is associated with various immunodeficiency diseases.
Defects in the CD73 gene can lead to calcification of joints and arteries, and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
