Product Specific Information

Description:
eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity. Expression on myeloid cells (CD11b) is restricted to the bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in mediating activation signals. Differences between human and mouse CD73 have been reported. In BALB/c mice, more CD4⁺CD73⁺ than CD8⁺CD73⁺ cells are observed, while the reciprocal is seen in humans.

CD73 is expressed on a subset of lymphocytes and increases during lymphocyte maturation. Memory CD4 T cells expressing CD73 resemble uncommitted primed precursor helper cells (Thpp) capable of differentiating into Th1 or Th2 cells. CD73 has also been found on regulatory T cells.

Applications Reported:
This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.2 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Optimize titration for best performance.

Kit Components:
Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Usage Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails before mixing with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures engineered for optimized fluorescence signatures with minimal spillover and spread.

Target Information

CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase):
Catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate. It is a dimer of two identical 70 kDa subunits bound externally to the plasma membrane via a GPI linkage. CD73 serves as a marker of lymphocyte differentiation, and deficiencies are associated with various immunodeficiency diseases. The gene is localized to 6q14–q21. Defects can lead to calcification of joints and arteries, and intestinal tuberculosis. Two transcript variants encoding different isoforms have been identified.
