Product Specific Information

eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
CD73 expression on myeloid cells (CD11b) is restricted to the bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling. Differences between human and mouse CD73 have been observed — BALB/c mice show more CD4+CD73+ than CD8+CD73+, while humans exhibit the opposite.

CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. It is found on memory CD4 T cells and regulatory T cells, associated with uncommitted primed precursor helper cells (Thpp) that can differentiate into Th1 or Th2 cells.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Tested on mouse splenocytes at ≤ 0.2 µg/test (defined as the amount to stain cells in 100 µL final volume). Cell number may vary between 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

NovaFluor dye compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
Recommended viability dyes: Invitrogen LIVE/DEAD Fixable Dead Cell stains.

CellBlox Plus Blocking Buffer:
Required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample before combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology based on nucleic acid dye structures, engineered for minimal spectral spillover and spread.

Target Information

CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, preferentially AMP.
It forms a dimer of two identical 70 kDa subunits anchored to the plasma membrane via GPI linkage, serving as a marker of lymphocyte differentiation.
Deficiency in CD73 is associated with various immunodeficiency diseases.
The CD73 gene is localized to chromosome 6q14–q21.
Defects in CD73 can lead to joint and arterial calcification, and intestinal tuberculosis.
Two transcript variants encoding different isoforms have been identified.
