Target Information
Beta-catenin is an adherens junction (AJ) protein involved in cell-cell adhesion and epithelial layer maintenance. It interacts with E-cadherin and alpha-catenin to anchor the E-cadherin complex to the actin cytoskeleton. Beta-catenin also associates with the adenomatous polyposis coli protein and microtubules, participating in signaling pathways such as Wnt/Wingless, tyrosine kinases, and phosphatases. These interactions regulate morphogenesis and cell proliferation.
Beta-catenin is expressed in various hair follicle cell types and plays roles in pilomatrixoma, medulloblastoma, colorectal, ovarian, and other cancers. In the nucleus, it co-activates Lef/Tcf transcription factors, stimulating genes like cyclin D and c-myc that drive cell proliferation. Its dysregulation is linked to tumorigenesis in multiple solid tumors including breast, colon, liver, lung, gastric, prostate, and skin cancers.
