Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily.
CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains (130 and 140 kDa) produced by alternative splicing. It is highly expressed on mature B cells and B-cell lymphomas.

The extracellular portion contains seven Ig-like domains capable of binding sialic acid-containing ligands on epithelial, endothelial, B, and T cells. The intracellular portion includes six tyrosine residues within ITIM and activation-like motifs that regulate B-cell receptor signaling.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Use 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Note: NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which improves staining performance.
Add 5 µL per sample to antibody cocktails or single-color controls (10³–10⁸ cells per 100 µL).

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed on B-lymphocytes.
It appears early in B-cell differentiation and is useful for phenotyping mature leukemias.
CD22 binds preferentially to α2,6-linked sialic acid and plays a regulatory role in B-cell receptor signaling through phosphorylation and interactions with Src family kinases and phosphatases.

Expression:

• Strongly expressed in hairy cell leukemia
• Positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma
• Negative in classical Hodgkin’s lymphoma
