Product Specific Information

Description:
The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a type I transmembrane glycoprotein belonging to the siglec subgroup of the Ig superfamily. CD22 is expressed at high levels on mature B cells and B cell lymphomas. The extracellular domain contains seven Ig-like domains that bind sialic acid moieties on epithelial, endothelial, B, and T cells. The intracellular portion includes six tyrosine residues within ITIM and activation-like motifs that regulate B-cell receptor signaling.

Each product includes:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:
Validated for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining with NovaFluor conjugates.
    • Add 5 µL per sample to antibody cocktails before mixing with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Based on Phiton™ technology utilizing nucleic acid dye structures engineered for optimal fluorescent signatures with reduced spectral spillover.

Target Information

CD22 (BL-CAM) is a 130–140 kDa type I integral membrane glycoprotein expressed on B-lymphocytes. Expression begins at the same differentiation stage as CD19 and is limited to mature B cells (CD22⁺) and absent in plasma cells (CD22⁻).
CD22 is strongly expressed in hairy cell leukemia and may assist in phenotyping mature leukemias. It binds preferentially to α2,6-linked sialic acid, and its phosphorylation is involved in regulation of B-cell receptor signaling. CD22 interacts with Src family kinases for positive regulation and recruits phosphatases for inhibitory signaling. It is also positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.
