Product Specific Information

The eBio4KB128 monoclonal antibody recognizes human CD22 (Siglec-2), a member of the siglec subgroup of the Ig superfamily. CD22 is a type I transmembrane glycoprotein composed of two polypeptide chains, CD22α and CD22β (130 and 140 kDa), produced by alternative splicing. It is highly expressed on mature B cells and B cell lymphomas. The extracellular region contains seven Ig-like domains that bind sialic acid-containing ligands, while the intracellular region includes tyrosine residues within ITIM and activation-like motifs involved in B-cell receptor signaling regulation.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.2 µg) per test. Each test corresponds to staining a 100 µL cell sample (10⁵–10⁸ cells/test).

Each product includes:

  • 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures that enable engineered fluorescent signatures optimized for spillover and spread.

Target Information

CD22 (BL-CAM) is a type I integral membrane glycoprotein (130–140 kDa) expressed in both the cytoplasm and membrane of B-lymphocytes. It appears early in B-cell differentiation, similar to CD19. CD22 membrane expression is limited to mature B cells and is useful for phenotyping mature leukemias. It is strongly expressed in hairy cell leukemia and positive in diffuse large B-cell lymphoma and nodular lymphocyte predominance Hodgkin’s lymphoma, but negative in classical Hodgkin’s lymphoma.

CD22 binds preferentially to α2,6-linked sialic acid. Its phosphorylation during immune response regulates B-cell receptor signaling, acting both as a positive and negative regulator via interactions with Src family kinases and SH2-domain phosphatases.
