Product Specific Information

Description:
The O323 monoclonal antibody reacts with human CD27, a lymphocyte-specific member of the TNFR superfamily. CD27 is expressed by a subset of thymocytes and most mature T cells, and is upregulated upon T-cell stimulation. CD27 binds to CD70 and plays an important role in T cell–B cell interaction.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use at 5 µL (0.25 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in 100 µL final volume. Cell number can range from 10⁵ to 10⁸ cells/test.

Target Information

CD27 is a 50 kDa member of the TNF receptor superfamily, including CD40 and CD30. It regulates cell proliferation and death, and its expression is mainly confined to lymphocytes. High CD27 expression depends on antigen receptor ligation, while CD70 expression requires co-stimulatory or pro-inflammatory signals.

CD27 is expressed as a disulfide-linked homodimer on mature thymocytes, peripheral blood T cells, and a subset of B cells. T cell activation upregulates CD27 expression and induces release of soluble CD27 (sCD27), a prognostic marker in B cell malignancies.

RgpA, a cysteine proteinase, can degrade CD27 and counteract CD27-mediated T cell activation. CD27-binding protein (SIVA) is a proapoptotic protein involved in CD27-induced apoptosis.
