Product Specific Information

The JJ319 monoclonal antibody reacts with the rat CD28 molecule, expressed by thymocytes, mature T cells, and a subset of NK cells. CD28 serves as a ligand for CD80 (B7-1) and CD86 (B7-2), acting as a potent costimulator of T cells. Signaling through CD28 augments IL-2 and IL-2 receptor expression as well as cytotoxicity of CD3-activated T cells. No reactivity of JJ319 is observed on mouse T cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of rat splenocytes. Recommended ≤0.5 µg per test (100 µL final volume). Cell number may range from 10⁵–10⁸ cells/test. Antibody titration is recommended for optimal performance.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T cell–specific surface glycoprotein and a member of the immunoglobulin superfamily. It is constitutively expressed on most peripheral blood T lymphocytes and provides a critical costimulatory signal upon binding CD80/CD86 on antigen-presenting cells. CD28 prevents T cells from entering an anergic or apoptotic state. In mice, CD28 expression is found on all CD4⁺ and CD8⁺ T cells in peripheral lymphoid organs and blood, with higher expression on immature thymocytes. Dysregulation of CD28 is associated with diseases such as mycosis fungoides and Sézary’s disease.
