Product Specific Information

Description:
The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions. Commonly used as a phenotypic marker for mouse T cells.

Applications Reported:
Used for flow cytometric analysis. Internal testing shows NovaFluor Yellow 570 may non-specifically stain B cells in SJL mice, but not in BALB/c or C57BL/6 mice.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤1.0 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Compatibility Notes:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Blocking Buffer:
Each NovaFluor conjugate ships with CellBlox Blocking Buffer. Use 5 µL per sample (regardless of number of NovaFluors). For single-color controls, use 5 µL per 100 µL cell sample containing 10³–10⁸ cells.

NovaFluor conjugates are based on Phiton technology utilizing nucleic acid dye structures engineered for optimized fluorescent signatures and minimized spillover/spread.

Target Information

CD3 complex transduces antigen-recognition signals in T cells and regulates TCR surface expression. Subunits (gamma, delta, epsilon, zeta) are encoded on chromosome 11 and belong to the immunoglobulin superfamily. CD3 is expressed on 68–82% of peripheral blood lymphocytes and 65–85% of thymocytes, but not on B or NK cells. Defects in CD3 genes are linked to immunodeficiency.
