Product Specific Information

The 145-2C11 monoclonal antibody reacts with mouse CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells. Binding of 145-2C11 to TCR initiates intracellular signaling leading to activation, proliferation, or apoptosis depending on conditions.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis of mouse splenocytes
Applications Tested: ≤1.0 µg/test (100 µL final volume, 10⁵–10⁸ cells/test)
Antibody titration recommended for optimal performance.

Important Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Improved formulation over CellBlox
• Required for optimal staining
• Add 5 µL per sample before combining with cells
• For single-color controls: 5 µL per 100 µL cell sample (10³–10⁸ cells)

Target Information

CD3 complex transduces antigen-recognition signals and regulates TCR expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, members of the immunoglobulin superfamily encoded on chromosome 11. CD3 plays a role in TCR-induced growth arrest, survival, and proliferation. Expressed on 68–82% of peripheral lymphocytes and 65–85% of thymocytes; absent on B and NK cells. Defects in CD3 gene are associated with immunodeficiency.
