Target Information
The tachykinins are a family of small peptides including substance P, neurokinin A, and neurokinin B. These peptides act on three distinct seven-transmembrane G-protein-coupled receptors at different concentrations.
The NK-3 receptor (NK3R) has the highest affinity for neurokinin B and is highly expressed in the supraoptic and paraventricular nuclei. Upon ligand binding, NK3R activates a phosphatidylinositol-calcium second messenger system, likely leading to vasopressin and oxytocin release.
NK3R may play a role in learning and memory, as mice lacking this gene show cognitive deficits. It may also be involved in vagal afferent neuron regulation, although activation by substance P or neurokinin A is more likely, since airway nerves do not express neurokinin B.
