Target Information
ICAD는 caspase-3-activated DNase(CAD)의 억제 단백질로, 핵 내 세포사멸(apoptosis)을 조절하는 기질입니다. ICAD는 두 가지 isoform을 가지며, 45kDa의 ICAD-L(DFF45)와 35kDa의 ICAD-S(DFF35)가 존재합니다. 두 isoform 모두 CAD를 결합하고 억제할 수 있지만, ICAD-L만이 기능적으로 활성화된 것으로 알려져 있습니다. ICAD는 caspase-3에 의해 절단되어 CAD가 핵 내 DNA 절단을 수행할 수 있도록 활성화됩니다. 또한 ICAD는 CAD의 특이적 접힘을 돕는 chaperone 역할을 합니다. 최근 연구에 따르면 ICAD는 대장암 및 뇌암 세포에서 정상적인 세포사멸 신호전달 경로를 회복시키는 잠재적 타깃으로 보고되고 있습니다.
