Target Information
Specialized cells at the midline, which separates the left and right halves of the CNS, have roles in directing growth cone behavior. In vertebrate spinal cord, insect ventral nerve cord, and C. elegans, midline cells produce guidance cues such as nectins and slit, acting as attractants and repellents. Robo functions as a receptor for the repellent Slit and defines a subfamily of Ig superfamily proteins conserved from fruit flies to mammals. Non-crossing axons express high levels of Robo, while crossing axons express low levels before reaching the midline and high levels after crossing. Robo1 and Robo2 are human homologs of the Drosophila Roundabout protein. Robo1 is homologous to the C. elegans gene sax3; Robo2 to the zebrafish gene astray.
