Target Information

FGFR3 (Fibroblast Growth Factor Receptor 3) is a member of the FGFR family of receptor tyrosine kinases. This family regulates various cellular functions including angiogenesis, mitogenesis, osteogenesis, myogenesis, carcinogenesis, cellular differentiation, and tissue repair after injury. FGFR family members are implicated in diseases such as cancer, rheumatoid arthritis, and diabetic retinopathy.

FGFR family members differ in ligand affinities and tissue distribution. A full-length FGFR3 protein includes:

  • Three immunoglobulin-like extracellular domains
  • A single hydrophobic membrane-spanning segment
  • A cytoplasmic tyrosine kinase domain

The extracellular region binds fibroblast growth factors, triggering downstream signaling that influences mitogenesis and differentiation. FGFR3 plays a critical role in bone development and maintenance. Mutations in this gene are associated with craniosynostosis and various skeletal dysplasias. Three alternatively spliced transcript variants encoding different isoforms have been identified.
