Product Specific Information

Description:
The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except plasma cells. Follicular dendritic cells also express CD19. CD19 forms a complex with CD21, CD81, Leu13, and MHC class II, associating with the BCR to mediate signaling, tyrosine phosphorylation, calcium flux, and B cell proliferation. The HIB19 and SJ25C1 antibodies recognize overlapping epitopes.

• Flow cytometric analysis.

Applications Tested:

• Pre-titrated and validated for flow cytometry using normal human peripheral blood cells.
• Recommended use: 5 µL (0.25 µg) per test (100 µL final volume).
• Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity:

• Tandem dye is sensitive to photo-induced oxidation; protect from light.

Fixation:

• Samples can be stored in IC Fixation Buffer (Product #00-822-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
• Clone-specific performance should be tested empirically.

Excitation/Emission:

• Excitation: 488–561 nm

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on all peripheral B cells and certain myeloid leukemia cells. CD19 serves as a functional regulator of B cell proliferation and differentiation and is expressed in all B cell precursor leukemias. It lowers the threshold for BCR-dependent stimulation and acts as a signal-amplifying co-receptor. Mutations cause hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
