Target Information

Epithelial sodium channels (ENaC) are amiloride-sensitive members of the degenerin/epithelial sodium channel (Deg/ENaC) superfamily of ion channels. These channels share structural similarities, including two short intracellular termini, two membrane-spanning segments, and a large extracellular loop with a conserved cysteine-rich region.

ENaC has three homologous isoforms (α, β, γ) and plays a key role in sodium and fluid balance in the kidney, lung, and colon. It also mediates aldosterone-dependent sodium reabsorption in the distal nephron, contributing to blood pressure regulation.

Mutations in ENaC subunits can cause conditions such as Liddle’s syndrome (gain-of-function in β or γ subunits) or pseudohypoaldosteronism type 1 (loss-of-function in α or β subunits). ENaC activity is also influenced by interaction with the CFTR chloride channel.
