Target Information

HEF-1 (also known as NEDD9) is a multifunctional protein involved in integrin-based signaling pathways that regulate cell motility, growth, apoptosis, and oncogenic transformation.
The Cas family of docking proteins, including HEF1, are key substrates of focal adhesion kinase (FAK) and Src family tyrosine kinases, both of which are significant drug development targets.
HEF1 overexpression enhances transcription of genes associated with motility and invasiveness, such as metalloproteinases, MLCK, p160ROCK, and ErbB1, and can mediate apoptosis in epithelial-derived cell lines (e.g., MCF7, HeLa).
Overexpression of BCAR1 (p130Cas), a related protein, has been linked to tamoxifen resistance, highlighting the importance of this protein family in cancer prognosis.
