Target Information
Caspase 9 (ICE-like apoptotic protease 6, ICE LAP6, apoptotic protease Mch6, apoptotic protease activating factor 3, Apaf3)는 CARD 도메인을 포함하는 peptidase family C14의 구성원입니다. Caspase들은 apoptosis를 매개하는 cysteine protease로, 비활성 전구체 형태로 발현되어 특정 기질을 절단함으로써 활성화됩니다. Caspase 9은 heterotetramer 형태로 활성화되며, 세포질에 존재하다가 활성화 시 미토콘드리아로 이동합니다.
Caspase 9은 apoptosis 실행에 관여하는 caspase 활성화 연쇄 반응의 핵심 효소로, Caspase 3 및 Caspase 6을 절단 및 활성화합니다. Apaf-1과 결합 시 protease 활성화가 유도되어 caspase-3을 절단하고 DNA 손상 유도 apoptosis를 촉진합니다. 또한 poly(ADP-ribose) polymerase (PARP)를 절단하며, 다양한 인간 조직에서 발현됩니다.
쥐에서 Caspase-9의 isoform이 확인되었으며, 이는 C 말단이 대체 서열로 치환된 형태입니다. Caspase 9의 돌연변이는 뇌 발달에 영향을 미치며, 부족한 Caspase 9은 비정상적인 대뇌 및 신경 발달을 초래합니다. 인간에서는 Caspase 9 발현 이상이 조직별로 다르며, 낮은 수준은 암 및 알츠하이머병과 같은 신경퇴행성 질환 발병에 관여할 수 있습니다.
