Product Specific Information

Description:
The 2E7 monoclonal antibody reacts with mouse CD103, the alphaE integrin. CD103 non-covalently associates with integrin beta7 and is expressed on intraepithelial lymphocytes, a subset of peripheral lymphocytes, and dendritic epidermal T cells (DEC). Epithelial cell antigen E-cadherin binds to CD103 and mediates lymphocyte homing to the intestinal epithelium.

Applications Reported:
Reported for use in flow cytometric analysis and immunohistochemical staining of frozen tissue sections.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Cell number can range from 10⁵–10⁸ cells/test. Antibody should be titrated for optimal performance.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated cells, including a subset of TGF-β1 cells. It consists of a 150 kDa alpha chain and a 120 kDa beta chain. CD103 is expressed by 0.5–2% of resting lymphocytes in blood and lymphoid organs. It stains few bone marrow cells and rarely peripheral lymphocytes. As part of the integrin family, CD103 forms a heterodimeric complex with beta7 integrin to create the E-cadherin binding receptor known as human mucosal lymphocyte-1 antigen. It is preferentially expressed in intestinal intraepithelial lymphocytes (IEL), contributing to adhesion and IEL activation. CD103 dysfunction is associated with hairy cell leukemia and splenic marginal zone lymphoma.
