Target Information
Ubiquitin exists either covalently attached to another protein or free (unanchored). When covalently bound, it is conjugated to target proteins via an isopeptide bond as monoubiquitin, polyubiquitin chains, or linear polymers.
Polyubiquitin chains have distinct biological roles depending on the lysine linkage type:
- Lys-6-linked: DNA repair
- Lys-11-linked: ERAD and cell-cycle regulation
- Lys-29-linked: Lysosomal degradation
- Lys-33-linked: Kinase modification
- Lys-48-linked: Proteasomal degradation
- Lys-63-linked: Endocytosis, DNA-damage response, and NF-κB activation
Linear polymers (Met-linked) are involved in cell signaling.
Free polyubiquitin also functions in kinase activation and signaling.
